RNA polymerase is responsible for one of the key cellular mechanisms.
It transcribes the genetic information stored in DNA into RNA. This
project focuses on a characterization of dynamical parts of the RNA
polymerase that are specific for Gram positive bacteria (including
serious pathogens) and exhibit conformational motions potentially
important for the biological function. Dynamics of two components of RNA polymerase holoenzyme of a model Gram positive bacterium Bacillus
subtilis has been studied so far, showing that they exhibit motions
that could be important for the function of RNA polymerase, but a detailed study is missing. This project will describe the
conformational exchange of these parts using advanced methods of
nuclear magnetic resonance (NMR). The project is expected to
contribute to the effort to describe the functional properties of the
highly flexible C-terminal domain of delta subunit of the RNA polymerase and understanding its role within the whole RNA polymerase by a detailed analysis of its fast dynamics.
